Overview

Glutathione (GSH) is a tripeptide composed of glutamate, cysteine, and glycine, joined by an unusual gamma-peptide bond. It is the most abundant intracellular antioxidant in the human body, present in virtually every cell at millimolar concentrations. Glutathione plays a central role in antioxidant defense, detoxification (Phase II conjugation), immune function, and cellular redox signaling. Levels decline significantly with age, chronic illness, toxin exposure, and oxidative stress, which has made glutathione supplementation and repletion a major focus of anti-aging and integrative medicine research. Mechanism of Action

Glutathione functions through multiple interconnected mechanisms. As a direct antioxidant, it neutralizes reactive oxygen species (ROS) and reactive nitrogen species (RNS), protecting cellular structures from oxidative damage. As a cofactor for glutathione peroxidase and glutathione S-transferase enzymes, it catalyzes the detoxification of peroxides, electrophiles, and xenobiotics (Phase II detoxification). Glutathione maintains the reduced state of other antioxidants including vitamins C and E, recycling them back to their active forms. It regulates the cellular redox state (GSH/GSSG ratio), which acts as a master switch for gene expression, immune cell activation, and apoptotic signaling. In immune function, glutathione is essential for lymphocyte proliferation, natural killer cell activity, and T-cell function.
