Overview

PE-22-28 is a heptapeptide fragment derived from proenkephalin that acts as a selective sigma-1 receptor agonist. The sigma-1 receptor is an intracellular chaperone protein expressed throughout the central nervous system that modulates calcium signaling, ER stress responses, and neurotrophic factor expression. PE-22-28 has gained attention in the nootropic and cognitive enhancement research community for its ability to promote neuroregeneration, upregulate BDNF, and demonstrate antidepressant-like effects in preclinical models. Unlike many sigma receptor ligands that are small molecules, PE-22-28 is a peptide-based agonist, which may confer different pharmacokinetic and selectivity properties. Research is still in early stages, with most published data coming from in vitro and animal studies.

Mechanism of Action

PE-22-28 binds to and activates the sigma-1 receptor, which acts as a molecular chaperone at the mitochondria-associated ER membrane (MAM). Activation promotes several downstream effects: enhanced BDNF release supporting neuroplasticity and neuronal survival, modulation of calcium signaling important for synaptic function, protection against ER stress-induced apoptosis, and potentiation of NMDA receptor-mediated neurotransmission involved in learning and memory. The sigma-1 receptor also regulates ion channel activity and dopamine/serotonin signaling, which may underlie the antidepressant-like behavioral effects observed in animal models.

Research Applications

Neuroregeneration research, cognitive enhancement and nootropic studies, BDNF upregulation, antidepressant mechanism research, sigma-1 receptor pharmacology, neuroprotection studies, synaptic plasticity research
